The KOH (Potassium Hydroxide) preparation is a “clearing” technique. Sputum is naturally thick and full of human cells and debris that can hide tiny fungi. When KOH is added to the sample, it dissolves the human cells and mucus but leaves the fungal cell walls intact because they are made of a tough substance called chitin.
