Mycobacterium tuberculosis has a unique, waxy cell wall that resists standard staining. The AFB Stain (Ziehl-Neelsen or Kinyoun method) uses a bright red dye that penetrates this wax. Once stained, the bacteria resist “decolorization” by acid-alcohol, remaining bright red against a blue or green background under a microscope.
